Osimertinib Plus Platinum-Pemetrexed: A Review in Previously Untreated, EGFR-Mutated, Locally Advanced or Metastatic NSCLC.

Oral osimertinib (TAGRISSO®) is a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. It is approved in several countries (including China, EU, Japan and the USA) for use in combination with platinum-pemetrexed chemotherapy as first-line treatment for adult patients with advanced non-small cell lung cancer (NSCLC) (EU) or locally advanced or metastatic NSCLC (USA) whose tumours have EGFR exon 19 deletions or exon 21 L858R mutations. In the pivotal phase III FLAURA2 trial in this patient population, osimertinib plus platinum-pemetrexed significantly prolonged median progression-free survival (PFS) and overall survival (OS) compared with osimertinib monotherapy. Osimertinib plus platinum-pemetrexed had a manageable tolerability profile, with safety consistent with that of the individual agents, and no new safety signals were identified. In conclusion, osimertinib plus platinum-pemetrexed is a valuable addition to the first-line treatment options for EGFR-mutated, locally advanced or metastatic NSCLC.
