2O Osimertinib (osi) with or without chemotherapy (CTx) as first-line treatment in EGFR-mutant (EGFRm) advanced NSCLC with concurrent TP53 mutations (TOP study)

Concurrent TP53 mutations are common in EGFRm NSCLC and associated with poor outcomes to EGFR-TKIs monotherapy. While the FLAURA2 study demonstrated superior efficacy of osi plus CTx in advanced EGFRm NSCLC, whether those with TP53 co-mutations could derive more benefit from combination therapy remains unclear. This randomized, multicenter, phase III trial addresses this critical gap by assessing the efficacy of combination therapy in this high-risk population.
