IN SILICO STUDY ON PEDIATRIC HIGH-GRADE GLIOMAS

Pediatric High-Grade Gliomas (pHGGs) encompass a group of emerging glial cell tumors characterized by their rare and aggressive nature with unpromising therapeutic prospects. Studies on pediatric glial tumors have revealed specific mutations related to histone variants, TP53 (Tumor Protein 53), and growth factors, differing from adult tumors that are widely mutated in IDH (Isocitrate Dehydrogenase gene). Within this framework, it is possible to suggest, given the unique biology of these tumors that sets them apart from High-Grade Gliomas in adults, the need for the development of targeted and specific therapies for pediatric patients. To contribute to the emerging studies on pHGGs, this research aimed to unveil in a more specific manner the protein interactions that characterize these tumors. Computational Biology tools were used for the construction and analysis of a Protein-Protein Interaction Network to elucidate tumor characteristics and analyze important interactions for the development of therapeutic targets. Through the in silico approach, it was possible to conclude the importance of studying central proteins such as VEGFA (Vascular Endothelial Growth Factor A), p53 (Tumor Protein 53), EP300 (Histone Acetyltransferase p300), and CREBBP (CREB-Binding Protein) in the organization of the tumor and to assess, from a general perspective, that these proteins may act as potential therapeutic targets. Thus, the advancement of Computational Biology, Biotechnology, and Systems Biology in partnership with Pharmacogenomics was found to be essential, as these tools provide more refined analyses of tumor characteristics that are difficult to describe in vitro, guiding future studies.
