This study examined how meloxicam (MEL) affects liver and subcutaneous adipose transcriptomes in periparturient obese dairy cows. Prepartum MEL administration 14 days before calving reduced inflammatory diseases, including mastitis, metritis, and endometritis, and improved reproductive performance. Transcriptomic analysis showed that MEL suppressed hepatic inflammatory processes while enhancing innate immune functions in adipose tissue. These findings indicate that MEL alleviates immune suppression and systemic inflammation associated with negative energy balance, thereby lowering disease risk and improving reproductive efficiency, and provide a theoretical basis for optimizing nutritional regulation and health management during the transition period. Supplementary files contain all serum datasets and complete transcriptomic analysis results generated in this study.
