Role of CDC25B in human cancers: a meta-analysis and bioinformatics analysis

CDC25B, as an important member of the CDC25 family, plays a vital role in cell regulation. Recent studies have identified it as a potential oncogene implicated in tumorigenesis and cancer progression. The goal of our research was to shed light on the usefulness and significance of CDC25B as a reliable marker for predicting cancer outcomes. The databases PubMed, Web of Science, China National Knowledge Infrastructure (CNKI) and Wanfang data were used to find relevant articles published prior to May 30, 2025. The correlation between CDC25B expression and tumor prognosis or clinical features by calculating the overall hazard ratio or odds ratio and 95% confidence interval. To further verify our results, we also employed an online database from The Cancer Genome Atlas (TCGA) and GTEx. Our study selected 16 articles for meta-analysis, including 2056 cancer patients. The current study showed that higher CDC25B expression was linked to poorer overall survival (OS) (HR =1.90, 95% CI: 1.45–2.48) and disease free survival (DFS) (HR =5.99, 95% CI: 1.07–33.63) in some cancers. Subgroups analysis indicated that CDC25B was significantly linked to poor OS in digestive system cancer, breast cancer and other tumors, but not related to repiratory system cancer. Correlation analysis demonstrated that CDC25B was linked to lymph node metastasis, T stage and TNM stage, not related to gender and clinical stage. Sensitive analysis indicated that the analysis of OS and T stage was stable. Begg’s and Egger’s tests indicated no publication bias in terms of gender, lymph node metastasis, T stage, and TNM stage. Bioinformatics analysis showed that CDC25B was over-expressed in most cancers. Cox regression found that CDC25B expression was linked to OS, DSS and PFI in ACC, KIRC, LIHC, and UVM. The frequency of CDC25B gene alteration was highest in OV. GSEA analysis demonstrated that CDC25B was invoved in apoptosis and cell cycle in most tumors. CDC25B was correlated with immnue cell infiltration in cancers. Abnormal CDC25B expression is linked to the clinical prognosis and pathological characteristics of different cancers. CDC25B expression levels are only associated with prognosis in specific cancer types (KIRC, LIHC, UVM).
