Pain pharmacology encompasses a complex relationship between neurobiological pathways, receptor systems, and pharmacologic agents that mediate or modulate pain perception and inflammation. In the context of the ongoing opioid crisis, understanding traditional and emerging approaches to pain management is critical. This educational review provides an integrative overview of pharmacologic strategies for acute and chronic pain, examining mechanisms of action, therapeutic applications, and safety considerations, across diverse drug classes including non-opioid analgesics, anti-inflammatory agents, muscle relaxants, and opioids. By synthesizing current evidence on the efficacy and safety of different treatments, this review highlights the evolving pharmacological basis for our clinical treatment strategies in pain management.
