Combining monoclonal antibodies with chemoradiotherapy is effective and safe for LACC, demonstrating favorable treatment outcomes.
PURPOSE: This study aimed to systematically evaluate the efficacy and safety of combining monoclonal antibodies with chemoradiotherapy for the treatment of locally advanced cervical cancer (LACC) in Chinese patients.
MATERIALS AND METHODS: A systematic search was performed in PubMed, Web of Science, Excerpta Medica Database (Embase), China National Knowledge Infrastructure (CNKI), and WanFang for randomized controlled trials, cohort studies, and prospective single-arm studies assessing monoclonal antibodies (anti-EGFR, anti-VEGF, and PD-1 inhibitors) in combination with chemoradiotherapy for LACC, from inception to 5 February 2025. Literature screening, data extraction, and bias assessment were conducted independently by two researchers. Meta-analysis was performed using Stata version 14.0 software.
RESULTS: Eighteen studies involving 776 patients were included. The pooled objective response rate was 90% (95% confidence interval [CI]: 0.85-0.95), complete response rate 65% (95% CI: 0.56-0.74), and disease control rate 94% (95% CI: 0.92-0.96). The 1-, 2-, 3-, and 5-year overall survival rates were 98%, 93%, 89%, and 80%, respectively. Common adverse events included gastrointestinal reactions (55%), leukopenia (51%), anemia (59%), and fatigue (20%). The mortality rate was 10%, and recurrence rate was 22% (95% CI: 0.12-0.35).
CONCLUSIONS: Combining monoclonal antibodies with chemoradiotherapy is effective and safe for LACC, demonstrating favorable treatment outcomes.
