We present Cy5Ql-SH, a water-soluble NIR probe with charge-switch-gated PET that reacts equally with Cys/Hcy/GSH for true total biothiol quantification. Using 20-200× diluted plasma, it measured total biothiols in healthy (189.2 μM) vs. liver-injured (122.1 μM) mice and was commercially validated.
