Although covalent organic frameworks (COF) have emerged as highly promising porous materials, yet their inherently poor aqueous dispersibility remains a critical bottleneck. Conventional dispersibility modification strategies often come at the cost of COF' intrinsic property while single-modal therapeutic approaches are insufficient to overcome tumor heterogeneity and robust intracellular redox defense systems. Herein, we report a replacement strategy that noncovalently functionalizes donor-acceptor conjugated COF with glucose oxidase. High photothermal conversion can be easily obtained, and such design simultaneously realizes long-standing aqueous dispersion with a low polydispersity index of 0.2. Subsequent deposition of gold nanoparticles and manganese dioxide enables synergistic dual-photothermal therapy, self-supplementing cascade chemodynamic therapy, and glutathione depletion. Detailed experimental results confirm that the generation of reactive oxygen species, fast catalysis of glucose, and decrease of mitochondrial membrane potential synergistically contribute to the excellent therapy effect.
