Here, we present a protocol for assessing endolysosomal chloride channel function in mammalian cells using the endolysosomal patch-clamp technique. We describe steps for vesicle enlargement, fabrication and fire-polishing of high-resistance pipettes, and mechanical dissection and isolation of enlarged vesicles.
