DNA modifications are central to carcinogenesis, chemotherapy drug action, neurodegeneration, and aging. Here, we present a protocol for click-code-seq, a next-generation sequencing technique for genome-wide single-nucleotide-resolution mapping of major types of DNA modifications: oxidized guanines and abasic sites. We describe steps for genomic DNA extraction and library preparation, including enzymatic modification conversion and click-chemistry-based tagging.
