• A Flag-tag-based CMC platform was developed to enable highly specific and sensitive screening of CCR1 receptor antagonists. • Magnolol and honokiol, two natural compounds derived from Magnolia officinalis, were identified as novel CCR1 antagonists. • Competitive binding of magnolol, honokiol, and BX471 to CCR1 was confirmed by a newly designed Nile Reda probe. • Both compounds showed potent anti-inflammatory effects in vitro/vivo, supporting therapeutic potential. • This study established a high-throughput CMC-based platform to identify bioactive components in traditional medicine.
